HILIC (Hydrophilic Interaction Chromatography) is a normal-phase mode separation and also a technique to separate highly hydrophilic, ionic, and polar compounds which cannot be separated in a reversed-phase mode. When separating sugars, amino acids, and nucleic acid bases, HILIC uses a mixture of organic solvent and buffer solution with a high concentration of organic solvent which can be favorable over an aqueous mobile phase in reversed-phase chromatography. The HILIC mobile phase gives better sensitivity for mass spectrometry detection because of the high concentration of organic solvent. The following will highlight separation using HILIC mode with the Sunshell HPLC columns.
