Ultra-Sensitive LC-MRM Based Method for Rapid and High-Throughput Screening of Sugar Phosphate Isomers in Cell Culture

I’d like to thank Dan Su and Fatemeh Mousavi, Ph.D. of HD Biosciences Co., Ltd for sharing their poster from ASMS 2024: “Ultra-Sensitive LC-MRM Based Method for Rapid and High-Throughput Screening of Sugar Phosphate Isomers in Cell Culture.” While we aren’t directly involved, I found their work especially interesting as we have a deep interest in HILIC Mode Chromatography. The following is directly highlighted from the poster below.

Overview:
A robust and ultra-sensitive LC-MS/MS method for high-throughput screening of sugar-phosphate isomers.

Key Highlights:

  • Sugar phosphate isomers are vital in metabolic pathways like glycolysis and energy storage.
  • Traditional LC-MS analysis is challenging due to poor retention and coelution.
  • Their method uses HILIC chromatography and MS/MS, offering improved selectivity and separation.
  • High sensitivity (LLOQ at 1 ng/mL), linearity (r > 0.98), and precision (CV < 15%) were achieved.
  • The technique is optimized for high-throughput screening of 120 drug candidates, demonstrating robustness and reproducibility.

This approach presents an advancement in the analysis of sugar phosphates.
